Since Marvel first released Iron Man in 2008, they have repeatedly said that "is all connected" . They know that they are capable of a world final power (by themselves) and that the bad ones can not obtain them by any necessary means.

That's the best explanation we get from the first 11 movies. The Big Bang happened and these "otherworldly" powers that should not and should not belong to any other part of the universe are compacted into 6 colored stones. Each stone has its own power and name: Space, Mind, Reality, Power, Soul and Time.

It is VERY important to note here that in BOTH instances, Adam Warlock played a crucial role in the death of Thanos. Okay, so we know where the stones are now (minus one) and we know what they all do, and now what? Well Thanos needs to pick them up and imbue his gauntlet with the power of the stones. From the published trailer, we know with certainty that it has Space and Power Stones.

Another fun fact! Actually, there are TWO gauntlets! The one can be seen very briefly in Thor, in the collection room below the throne room and it seems to have fake stones inserted to show, because who just wants to look at a simple gold glove? BORED.
